Purification of GPCRs via affinity tags can yield highly pure proteins, but due to the high flexibility of the proteins typically only a fraction of the entire preparation is active. This is often reflected in SEC spectra that show different stages of aggregation. Purifying the same GPCR on a ligand-based affinity column can yield an active, homogeneous preparation. Whenever a specific ligand for a GPCR of interest is known, we can attempt to couple it to one of our activated agaroses or magnetic beads to yield a purification matrix that is specific to the active conformation of this protein. One well-known example for such a ligand-based purification is Alprenolol Agarose, which binds to the beta 1 and beta 2 adrenergic receptors. The ligand was coupled via a hydrophilic linker to our highly crosslinked 6% agarose to minimize non-specific binding. This matrix is available off the shelf in mL quantities. Please contact us
