Membrane protein purification should be easy!
But in practice, membrane proteins require special attention, especially if you need to preserve native protein properties for downstream assays.
Top 3 bottlenecks in membrane protein purification
Manual
handling
- Protocols are long
- It’s error-prone and can be user-dependent
- Purifying many samples manually is not scalable
Optimising
conditions
- Each protein behaves differently
- Ideal conditions cannot be predicted - screening is essential
- This adds time and complexity to the purification stage of a project
Preserving native
protein behaviour
- Solubilisation and stabilisation agents are key for native protein prep
- Traditional protocols remove native lipid environment with detergents
- Identifying best solubilisation agent takes time
PlateX MP™ plates – a tool for easier and faster membrane protein purification
PlateX MP™ plates are a ready-to-run product, designed for use with liquid handling robots. The workflow combines two experimental processes in a single run:

Screening
For optimal solubilisation agents that preserve native protein properties

Purification
Of membrane proteins that delivers analytical-grade proteins in only 2 hour
What do PlateX MP™ users say
Artem Evdokimov
CSO at Orogen Therapeutics
“Cube’s polymer-based workflow for automated membrane protein purification is a game-changing innovation. Having personally purified multiple GPCRs and ion channels using detergent micelles and protein nanodiscs, I was struck by the speed and simplicity of purifying membrane proteins directly from resuspended biomass into polymer nanodiscs in an essentially hands-off manner. The ability to screen multiple polymers in parallel often enables the rapid generation of functionally relevant protein samples on the first attempt.”
How do PlateX MP™ plates perform
To evaluate performance of PlateX MP™ plates, we purified multiple membrane proteins and assessed their yield, purity and stability. We show data for two GPCRs, a gap junction protein and an ion channel. We also evaluated the latter for suitability for downstream cryo-EM analysis.

What do PlateX MP™ users need to do
The workflow includes 3 steps:

Step 1
Load the PlateX MP™ plate with cell lysate.

Step 2
Start the run on your liquid handling robot.

Step 3
Collect purified proteins in 2 hours.
How does the PlateX MP™ technology work
PlateX MP™ plates are pre-filled deep well plates that allow to screen for optimal solubilisation agents and purify membrane proteins, in a single run.

PlateX MP™ plates are ready-to-run
The plates include all required components for the workflow:
- Pre-filled with a workflow mix of components for the entire protocol
- Lyophilised components, stable at room temperature
- Validated protocols for Integra and Jena Analytik liquid handling robots
The ready-to-run plate format eliminates manual steps so you can walkaway and let the robot run the whole workflow.
‘2 in 1 workflow mix’ for minimal hands-on time
The pre-loaded workflow mix includes all components for screening of optimal solubilisation agents (copolymers) while running the purification on your robot. The PlateX MP™ plates include:
- 8 copolymers for identification of optimal solubilisation agents
- Magnetic beads for affinity tag-based purification (3 options available)
- Components for all buffers already pre-loaded
This ‘2 in 1 workflow’ setup reduces time-to-purified protein by eliminating separate optimisation experiments and manual handling.
Synthetic copolymers for native-like protein properties
The PlateX MP™ plates include 8 synthetic copolymers from the Cubipol product range. Cubipols are next-generation copolymers that solubilise and stabilise membrane proteins in the form of nanodiscs.
- 8 Cubipol compounds for a full spectrum of copolymer properties
- Purified membrane proteins keep their native lipid environment
- Identify optimal copolymers for your protein in a single run
Copolymers preserve native membrane protein properties and are suitable for downstream assays such as drug screening or structural studies.
