Description

We are excited to introduce our off-the-shelf, full-length, active, and native Endosomal/lysosomal proton channel TMEM175, commonly known as TMEM175. The physiological role of TMEM175 is centered on maintaining the electrochemical gradient required for lysosomal catabolism.  Through its influence on the lysosomal membrane potential, TMEM175 indirectly modulates the efficiency of proton import. Loss of TMEM175 leads to lysosomal instability, resulting in either luminal over-acidification or impaired acidification depending on the metabolic state of the cell.  Genetic studies have identified common variants and rare loss-of-function mutations in TMEM175 that significantly increase the risk of developing Parkinson's Disease.

Function TMEM175 is a constitutively active lysosomal potassium (K+) channel that regulates organellar membrane potential and luminal pH stability, facilitating optimal enzymatic activity and autophagosome-lysosome fusion. It acts as a critical ion flux regulator to prevent lysosomal hyperpolarization during proton import by the v-ATPase.
