Description
NativeMP™ Strep-Tactin®XT Purification plates are 96 deep-well plates for high-throughput membrane protein purification of Strep-tag®II or Twin-Strep-tag® fusion proteins with KingFisher instruments. They contain MagStrep® Strep-Tactin®XT beads, which are characterized by a high binding specificity and affinity. The plates have to be combined with NativeMP™ Screening plates (Cat. No. 90302, 90304 or 90306) to identify optimal solubilization and stabilization conditions for a membrane protein of choice.

Quantitative analysis of membrane protein content and quality by SDS–PAGE, 330nm fluorescence and DLS. a.+ d. Solubilization and purification efficiency was screened for 14 MPs with 15 different NativeMPTM copolymers. Eluates were run an SDS–PAGEs (Coomassie stained). c. + e. Protein quantity was analyzed either by densitometry (ImageJ Gel Analyzer) or by buried tryptophan fluorescence at 330 nm (NanoTemper Panta Discovery). Intensities were normalized to the strongest signal in each dataset (set to 100%) and plotted as box plots to visualize distributions across copolymer families. d. For HTR7, eluates from all tested copolymers are shown (a), while for the remaining targets only the strongest eluates are displayed. Full, unmodified SDS– PAGEs and Western blots are provided in Fig. S1-3. In panel e, results from all 14 targets were combined to compare solubilization and stabilization efficiencies across the six copolymer backbones.
