MSP1D1-His "Ultra Dark" is a his-tagged membrane scaffold protein (MSP) derived from apolipoprotein A-I that wraps around a phospholipid bilayer patch to form stable, water-soluble nanodiscs, enabling detergent-free solubilization and stabilization of membrane proteins in a near-native lipid environment. Its defining feature is the replacement of all tryptophan and tyrosine residues with non-fluorescent side chains, resulting in an almost non-detectable UV absorbance at 280 nm, allowing for straightforward detection and accurate concentration determination of integrated membrane proteins within the disc.

Application The reduced UV absorption at 280 nm makes MSP1D1-His "Ultra Dark" particularly advantageous over standard MSPs and alternative solubilization agents such as AASTY or SMA, enabling reliable UV-Vis based concentration determination and significantly reduced background interference in fluorescence-based assays and label-free detection methods. Beyond its spectroscopic advantages, MSP1D1-His "Ultra Dark" is well suited for Cryo-EM, ELISA, and protein-protein interaction studies where unrestricted ligand access is required, and is further compatible with cell-free expression systems and a broad range of biomedical, proteomic, and protein biochemistry applications.

FAQ

Can I get the datasheet for the MSP1D1-His “Ultra Dark” protein?

Is this the correct MSP product for my membrane protein?

This cannot be answered easily as every membrane protein performs differently with each nanodisc size and phospholipid. It usually requires a screening process before. Remember that membrane proteins with fewer transmembrane domains (TMD) require a smaller nanodisc, while a high number of TMDs need a larger nanodisc.

Why are there different origins for the MSP protein and do they work differently?

First, no they all work the same. Do not worry. Second, the origin of the MSP protein is of importance e.g. western blots. As the secondary antibody is usually against a species e.g anti-mouse. To avoid unwanted cross reactions the origin of the MSP protein must be known.
